Title:	探討奈米膠體金對盤尼西林醯胺酵素之轉化效率影響(Investigation of nano-colloidal gold to the bioconversion efficiency of penicillin G acylase)

Abstract:	本研究應用奈米膠體金 (Colloidal Gold, CG)與盤尼醯林醯胺酵素 (penicillin Gacylase, PGA)，進行PG 生化反應生成 6-APA之轉化效率影響研究。首先製作不同粒徑之CG，利用其添加量、pH 值和溫度，探討上述因素對6-APA反應轉化效率之影響。實驗結果證實，添加 CG 可提高 PGA 對反應生成 6-APA 之轉化效率。其最佳添加條件為﹕PGA 加 0.05ml CG 溶液 (D=46.5nm)，在 pH8、37℃條件下，添加 CG 之轉化效率較未添加CG的增加2.13倍。另外，實驗發現CG效應為降低PG反應的活化能 (降低 11.7%)，使得生成 6-APA 之轉化效率提高。將 CG 經重複使用 7 次，仍維持生成6-APA 之轉化效率效果。
